靈芝(Ganoderma lucidum)在過去亞洲地區的傳統文化中就已經被當中藥材廣泛使用在治療或是預防疾病的發生,由於靈芝中的多醣體已被證實具有調節免疫力、抗發炎、抗氧化、抗癌和抗菌等功能,因此本試驗將探討靈芝水萃液作為肉雞抗發炎的飼料添加物之功效。本研究分為三個試驗,試驗一目的為測定靈芝的體外抗菌效果及生物活性成分的定量,試驗二是使用葡聚醣硫酸鈉(dextran sulfate sodium, DSS)作為肉雞促發炎藥物並測試靈芝水萃液對生長性能、腸道型態、抗發炎效果及腸道菌相的影響,試驗三使用脂多醣(lipopolysaccharide, LPS)誘導肉雞發炎並觀察靈芝水萃液對生長性

能、腸道型態、抗發炎效果及腸道菌相的功能。試驗結果顯示,靈芝水對金黃色葡萄球菌及大腸桿菌具有抑菌效果,此外,在兩種攻毒模式中靈芝水萃液都無法改善其生長性能,但在腸道的發炎基因(COX2、iNOS、IL-1β和IL-6) mRNA表現量皆有抑制的效果;在DSS攻毒的試驗中靈芝無法提高小腸的絨毛長度,但在LPS試驗中靈芝水萃液的高低劑量處理和對照組相比都顯著提高空腸和迴腸的絨毛長度;試驗二的腸道菌相豐富度會因為給予靈芝水萃液而顯著提高,並且抑制了變形菌門的比例,試驗三的結果則是可以發現給予高劑量靈芝水萃液處理會使alpha diversity中的simpson和shannon下降,腸道內乳酸桿菌的

數量相對上升,原本因為LPS攻毒而下降的糞桿菌也恢復成和對照組一樣的水平。綜合上述,靈芝水萃液無論是在DSS或是LPS攻毒皆具有抑制發炎反應的效果,並促進腸道內有益菌的含量,且幫助腸道菌群穩定,在LPS試驗中也能提高絨毛長度增加吸收面積,因此靈芝水萃液具有作為抗發炎飼料添加物的潛力。

肉雞養殖產業常面臨病原菌威脅、飼養空間限制、環境溫度過高及飼料汙染的挑戰,台灣飼料汙染又屬嘔吐毒素最為嚴重。在過去會以添加抗生素和傳統吸附劑來解決這些問題,然而抗生素會造成抗藥性細菌產生、殘留在食物中和環境汙染等問題,因此各國開始禁用抗生素;而傳統吸附劑的吸附能力不佳,並且有吸附飼料中微量元素的疑慮,需要另尋飼料添加物應對。精油對於肉雞擁有提升消化酵素、調節腸道菌群、抗氧化和免疫調節的功能;而芽孢桿菌發酵物在艱困環境中可形成孢子,更順利的到達腸道中定殖,且能分泌有抗菌能力的次級代謝物,在肉雞體內可調節細胞激素和腸道菌群,因此本實驗以精油與芽孢桿菌發酵物綜合添加,探討其對於肉雞生長性能、腸道型

態及盲腸菌相的影響。在體外抑菌試驗中,精油的添加對於枯草芽孢桿菌和地衣芽孢桿菌都無抑菌效果,適合綜合添加。而精油萃取液對於金黃色葡萄球菌和大腸桿菌都具有抑菌效果。在動物實驗一,證實芽孢桿菌發酵物可提升Lactobacillus菌屬相對含量;精油與芽孢桿菌發酵物綜合添加,則可提升Eubacterium hallii group和Christensenellaceae R 7 group菌屬相對含量,使其代謝物短鏈脂肪酸含量增加。精油則是透過提升抗氧化酶含量,來改善腸道絨毛型態。在動物實驗二,5 ppm嘔吐毒素攻毒模式下,精油與芽孢桿菌發酵物綜合添加可改善十二指腸段的絨毛高度與隱窩深度比值和顯著提

升空腸絨毛高度,並且提升菌群:Ruminococcaceae UCG 005、Ruminococcaceae UCG 014及Christensenellaceae R 7 group的相對含量,進而增加三甲基丁酸含量。產品吸附劑、降解劑及精油與芽孢桿菌發酵物綜合添加都可差異性調節促炎激素、抗氧化酶及緊密連接蛋白相關基因表現量。然而,只有精油與芽孢桿菌發酵物可提升肉雞血清中IFN-γ含量,使其與對照組無異。綜合上述,精油與芽孢桿菌發酵物的綜合添加,有能力調節腸道菌群及短鏈脂肪酸含量,進而改善腸道型態、促炎激素、抗氧化酶以及緊密連接蛋白相關基因表現量,具有取代抗生素的能力,且改善嘔吐毒素攻毒所造
